چکیده
Lebrikizumab (lebri) is a novel, monoclonal antibody that selectively targets IL-13 and prevents formation of the receptor alpha 1 (Rα1)/IL-4 (Rα) heterodimer signaling complex. A previous crystal structure report showed lebri does not interfere with binding to Rα2 decoy receptor. In contrast, other antibodies tralokinumab cendakimab had been reported inhibit both IL-13Rα1 IL-13Rα2. We wanted investigate whether interfered IL-13Rα2 subsequent internalization. Through competitive experiments using surface plasmon resonance, we confirmed can bind tralokinumab/IL-13 cendakimab/IL-13 complexes. These data binds at different epitope. First, induced expression on HaCaT cells, spontaneously transformed keratinocyte cell line from adult human skin, co-treatment TNF-alpha. Then, fluorescently stained these cells increased Using live-cell confocal imaging, observed internalized into cells. were conducted in presence an inhibitory ensure only through Importantly, also internalization IL-13/lebri complex, while IL-13/tralokinumab IL-13/cendakimab complexes do are The complex colocalized lysosome marker, indicating it likely be degraded lysosomes. summary, allows internalize This mode action differentiates cendakimab, since natural clearance levels

Enhanced Interleukin (IL)-13 Responses in Mice Lacking IL-13 Receptor α 2
Interleukin (IL)-13 has recently been shown to play important and unique roles in asthma, parasite immunity, and tumor recurrence. At least two distinct receptor components, IL-4 receptor (R)alpha and IL-13Ralpha1, mediate the diverse actions of IL-13. We have recently described an additional high affinity receptor for IL-13, IL-13Ralpha2, whose function in IL-13 signaling is unknown. To better...
متن کاملIL-13 receptor α2-arginase 2 pathway mediates IL-13-induced pulmonary hypertension.
Although previous literature suggests that interleukin (IL)-13, a T-helper type 2 cell effector cytokine, might be involved in the pathogenesis of pulmonary hypertension (PH), direct proof is lacking. Furthermore, a potential mechanism underlying IL-13-induced PH has never been explored. This study's goal was to investigate the role and mechanism of IL-13 in the pathogenesis of PH. Lung-specifi...
متن کاملIL-13R(alpha)2, a decoy receptor for IL-13 acts as an inhibitor of IL-4-dependent signal transduction in glioblastoma cells.
Interleukin (IL)-4 and IL-13 share the type II IL-4 receptor for cell signaling. We show that despite expressing the necessary signaling components, glioblastoma cells failed to respond to either IL-4 or IL-13. This was in part because of the expression of a high-affinity IL-13-binding transmembrane protein IL-13R(alpha)2 that inhibited IL-13-mediated Stat6 activation by acting as a decoy recep...
متن کاملIL-4 receptor alpha is an important modulator of IL-4 and IL-13 receptor binding: implications for the development of therapeutic targets.
IL-4 is a key cytokine associated with allergy and asthma. Induction of cell signaling by IL-4 involves interaction with its cognate receptors, a complex of IL-4Ralpha with either the common gamma-chain or the IL-13R chain alpha1 (IL-13Ralpha1). We found that IL-4 bound to the extracellular domain of IL-4Ralpha (soluble human (sh)IL-4Ralpha) with high affinity and specificity. In contrast with ...
